Umm your question is a bit confusing...
Do you mean why didn't they wake up in the van or in the airplane? I think the only persons that didn't wake up in the van were Cobb and Saito. But that's cause they were still in limbo, and heading on their way out.
If you mean why didn't they wake up in the plane immediately, that's because the people in the plane didn't have a "kick" to wake them up. While in the plane, they had to wait for the sedative to wear off before waking up. A bit confusing, but if you think through the levels, you should get it.
